Zone 4 route 41AF was an event route during the 2022 April Fool's event. Operated by all Sunshine Island operators, TRR, NWFB and KMB, the route travelled between Northern Beach and Sunshine Station. Unlike the standard route 41A, this route was serviced by buses converted to airplanes (with jet engines and wings), allowing them to fly over the Islands at 200 km/h.

Route 41AF was run as an express service, bypassing all intermediate stops while traveling to Sunshine Station and only making an intermediate stop at Western Harbour Tunnel Interchange on the return trip.
Vehicles "fly" by travelling on a hidden track that lies above Zone 4 and also drive through the pedestrian bridges near Central West on Northern Beach-bound trips. In addition, runways were built at Western Harbour Tunnel (on the Zone 4 side) and Northern Beach, as well as near Sunshine Square to allow vehicles enough room to take off and land. Invisible launch ramps could be found past Sunshine Station and the Zone 4 portal of the Western Harbour Tunnel. These have since been removed following the end of the April Fools' event. However, buses bound for Sunshine Station would land on South Central Road, owing to the lack of space for a dedicated runway near Southern Interchange, where they would drive in normal traffic.
Because of the specialized vehicles used, this service was only run when required by passenger demand. It also has the most expensive fare in the Islands. Despite this, with the jet engines and extremely quick travel time, this route saw very high ridership, with long lines at each stop.
The route also attracted some YouTubers to the Islands, which showed off the capabilities of the service.
After the event, the route remained drivable but as the express variant of route 41A, which required vehicles to be driven on the roads, adding a round trip through the Western Harbour Tunnel. This also caused ridership to drop as it took about the same time as the 41A. The route is no longer drivable as of the V2 update in June 2024.

- On 1 April 2022, as part of the April Fool's event, all Sunshine Islands operators (except Forever Transit), TRR, NWFB and KMB began temporary service on this route.
- On 3 April 2022, Forever Transit also temporarily operated this route.
- On 29 April 2022, with the end of the April Fool's event, service was officially discontinued and reverted to the standard route 41A. At the same, all jet buses were removed from 41AF service, although some could be driven in conventional service until May 31, 2024.

- This route was the only fixed route without any fixed timetable and was serviced by every operator in the game, irrespective of zones. Today, the only route serviced by all operators are Private Hire trips.
- Unlike Sunshine Stadium routes, this service solely existed for the 2022 April Fool's Event meme buses. Regular buses initially could not operate this route as they would fall out of the sky.
- The CSB Dennis Dart will show a blank display if route 41AF is selected (the rollsign does not have the exposure).
- According to the route menu, this service takes 2 minutes for a single trip, which made it the fastest route in Sunshine Islands.
- KMB bus stop flags were used for this route until April 29, 2022, then the route was made hail and ride.
- If you spawned a jet bus for use on conventional service, wings would not appear (although acceleration remained the same).
- For some reason, 41AF was still drivable until the end of V1. This could be done by selecting the special tab, then clicking 41AFS or 41AFN. Stops remained the same, however, service was done on the roads instead of in the sky. Effective April 29, 2022, all Northern Beach-bound trips made a roundtrip through the Western Harbour Tunnel but did not service stops in Zone 1 or the Zone 1-bound platform at the Western Harbour Tunnel Interchange.
- The Hitachi 5000A, which used a single cab car to make it a bus was temporarily added to the game specifically for this service. Horizon Group also tested hovercraft technology on this route with a modified Enviro200.
- This bus route was the only route to have two suffixes (A and F).
